Essentially you have two teams, a mafia which knows all of its teammates and is aiming to gain a majority by forcing lynches on town players, and a town which doesn't know any of its teammates and is trying to figure out who's in the town and who's mafia. Game alternates between day phases, where people talk and discuss lynch candidates, and night phases, where usually (but not always) the town doesn't talk and the mafia chooses someone to kill; game ends once a win condition is met.

Oh no no, just mafia. I'm not realistically sure how I'd make it a diplo variant. It uses the same skills though, roughly speaking (albeit with different emphases).

Do you play with characters other than mafia? For a large group I add medics and cops.

I do! I'm going to decide on what roles go in based on how many people I get. For a first-time game I'm going to keep the roles relatively vanilla, I think (something like cop/doc/roleblocker).

Contingent on numbers, I'm planning on having one cop (town aligned, checks a player's alignment town/scum each night) and one doc (town aligned, picks a player to save from nightkills each night, may pick self), and giving the mafia a factional roleblock (so in addition to the factional kill, they pick a target, and the target can't use their actions for that night phase).

As for the hybrid, I think it's doable. Play on any board that fits the number of players. Define one small faction of Mafia. Win conditions of the game are not individual, but factional.

In other words, the larger faction of citizens (or whatever the vanilla role is called in your local variant) needs to guess who the bad guys are, and eliminate them in-game.

Although honestly, the bad guys have an enormous advantage, so probably should be minimal.

I'm out if this is on a diplomacy board, I played in the King/knight/rebel/traitor games and they sucked.

I could see it. If I had time to play a full-press game I'd sign up. As it stands I can still host it and get it set up, if there's enough interest.
